Abhi, If it is a new company, then your first priority will be to support hiring needs of your business managers. If it is an existing company, then you need to identify all those HR activities currently shraed by others , pool them and organise under your Head. This is just to start with.

Dear Professional,
First understand the business of the organization and the operating environment in which your business is going to be done.
Based on the inputs from your seniors (i suppose they are from line function), you can do following activities:
1.Prepare basic administrative policies related to code of conduct, office timings, breaks, leaves, holidays, tour & travel, Recruitment, Compensation, Grade structure, General Rules & Regulations, Confidential policies, Dos & Donts., List of Holidays, Compensatory Offs, Various Formats, HR Mannual etc.
2. Prepare Authority matrix, Values, Vision & Mission Statements, Competency Matrix, Service Level Agreements, Induction Program, Goals, Objectives, Performance Parameters, Job Profiles etc.
3. Seek advice from Layers for all formats especially you will be exchangin with employees.
4. Get yourself clear about the expectation from Managements. Define your goals with proper time-lines.

The above is preliminary you have to do.
